蜂王浆对低温保存绵羊精子质量和糖转运蛋白GLUTs表达的影响
Effects of Royal Jelly Supplementation on Sheep Sperm Quality During Liquid Storage at 5℃

摘要(Abstract):

实验旨在研究稀释液中添加蜂王浆对低温保存绵羊精子质量和糖转运蛋白丰度的影响。通过假阴道法采集8只杜泊公羊精液,将活率不低于0.8的精液混合,分别用含0.00%、0.25%、0.50%、0.75%和1.00%蜂王浆的稀释液将精液稀释至8倍,0.00%蜂王浆组为对照组,置于5℃保存。在保存0、24、48、72 h利用计算机辅助精子分析系统、低渗膨胀实验、双荧光探针和Western Blot分别检测精子运动参数、质膜完整性、线粒体活性以及糖转运蛋白GLUT 3和GLUT 8丰度。结果表明:在低温保存72 h时,0.75%蜂王浆组精子的活率、活力、直线运动速率、路径速度、曲线运动速率、质膜完整性和线粒体活性均显著高于对照组。WesternBlot结果显示精子GLUT3和GLUT8丰度均随保存时间延长而降低,保存72h时GLUT8的丰度显著低于0 h;保存72 h时0.75%蜂王浆组GLUT 3和GLUT 8丰度显著高于对照组。综上,在稀释液中添加0.75%的蜂王浆可有效提高低温保存绵羊精子的质量,这与精子GLUTs蛋白表达和精子能量代谢密切相关。
This experiment was conducted to study the effect of royal jelly supplementation in diluent on sheep sperm quality and the expression of GLUTs during liquid storage at 5℃.The semen was collected by artificial vagina method from 8 Dorper ram.The mixed semen with the motility no less than 0.8 was diluted to 8 times using the semen extender containing 0,0.25%,0.50%,0.75% and 1.00% royal jelly.Five groups were stored at 5℃ for 0,24,48 h and 72 h.Parameters of sperm motility,plasma membrane integrity,mitochondrial activity and expression of GLUT 3 and GLUT 8 were respectively assessed by using computer-assisted sperm analysis(CASA),Hypoosmotic Swelling Test(HOST),JC-1 and PI double standard fluorescence staining and Western Blot.The results showed that when stored for 72 h,the sperm motility,sperm viability,VAP,VCL,VSL,plasma membrane integrity and mitochondrial activity of the 0.75% group were significantly higher than that of the control group(P<0.05).Western Blot results showed that the abundance of GLUT 3 and GLUT 8 was decreased with the extension of time,and the abundance of GLUT 8 was significantly higher at 0 h than that at 72 h(P<0.05);The 0.75% of royal jelly was added and storage after 72 h,the abundance of GLUT 3 and GLUT 8 was significantly higher than that of the control group(P<0.05).In summary,the semen extender supplement with 0.75% royal jelly could significantly improve sperm quality after storage,and it is closely related to the abundance of GLUTs and sperm energy metabolism.
