香炉山鸡隐性白羽鉴定分析Identification and Analysis of Recessive White Feather in Xianglushan Chicken

摘要(Abstract):
为淘汰香炉山鸡群体中的白羽基因杂合子个体,建立白羽基因纯合群体,本研究自行设计多重PCR扩增引物,采用PCR扩增结合琼脂糖凝胶电泳技术对120只香炉山鸡的基因型进行鉴定。结果显示:香炉山鸡隐性白羽CC型为56个,占总群体46.67%;Cc型为4个,占3.33%;cc为60个,占50%。经Popgene2.1软件统计分析发现隐性白羽基因位点平均等位基因数为2,有效等位基因数为1.9978,Shannon指数为0.692 6,多态信息含量为0.374 7,属于中度多态,说明该群体还具有一定的选育空间。本研究为香炉山鸡提供一种分子标记辅助选育方法,为该品种进一步的保护、开发与利用提供技术支撑,为其他鸡群选育研究提供参考。
In order to eliminate the hybrids in the Pheasant Group of Xianglushan chicken and establish two pure groups of Ma Yu and white Feather, this study designed the multiple PCR amplification primers, and the genotype of 120 Xianglushan chicken was identified by PCR amplification combined with agarose gel electrophoresis. The results showed that the recessive white feather CC type of Xianglushan chicken was 56, The total group 46.67%, Cc type 4, accounting for 3.33%, CC for 60, accounting for 50%; Through the statistical analysis of Popgene2.1 software, it was found that the average allele of gene loci of recessive white plume was 2, and the effective allele was 1.9978,Shannon index was 0.6926. The Polymorphism information content(PIC) was 0.3747, and the gene loci were moderate polymorphism, which indicates that the group also has a certain breeding space. This study provides a molecular marker assisted breeding method for Xianglushan chicken, which provides technical support for the further protection, development and utilization of the variety, and provides reference for other breeding studies of chickens with similar conditions.
