GDF9、BMP15基因在单、多羔黔北麻羊不同组织中的表达研究

摘要(Abstract):

为探讨GDF9基因和BMP15基因在单、多羔黔北麻羊性腺轴组织中的mRNA相对表达规律,本实验选取产单羔和多羔的经产母羊各3只,通过qPCR法对GDF9基因和BMP15基因在单、多羔黔北麻羊性腺组织中的相对表达水平进行检测。结果表明:GDF9、BMP15基因mRNA在单、多羔黔北麻羊5个性腺组织中均有不同程度的表达。由组间比较可知,多羔组的GDF9基因在垂体相对表达量极显著高于单羔组,在输卵管中单羔组的相对表达量显著高于多羔组,其他组织之间差异不显著;BMP15基因在输卵管中的表达量呈现为单羔组极显著高于多羔组,在子宫中呈现为单羔组显著高于多羔组,其余组织之间差异不显著。本实验结果表明GDF9基因和BMP15基因在黔北麻羊的单羔组和多羔组性腺轴组织之间的表达趋势大体一致,在卵巢中最高,在子宫中最低,说明2个基因对黔北麻羊的繁殖性能有重要作用。本研究结果可为今后深入探究GDF9基因和BMP15基因对山羊的繁殖机理提供理论依据。
In order to investigate the mRNA relative expression patterns of GDF9 and BMP15 genes in gonadal axis of single and multi-lambing Qianbei Ma goat,3 ewes were selected in this study,and qPCR method was used to detect the relative expression levels of GDF9 and BMP15 genes in gonadal tissues of single and multi-lambing Qianbei Ma goat.The results showed that GDF9 and BMP15 mRNA were expressed in five gonadal tissues of single and multi-lambing Qianbei Ma goat.The relative expression level of GDF9 gene in the pituitary gland of the multi-lambing group was significantly higher than that of the single lambing group,and the relative expression level of GDF9 gene in the fallopian tube of the single lambing group was significantly higher than that of the multi-lambing group,but there was no significant difference among other tissues.The expression level of BMP15 gene in the oviduct was significantly higher in the single lambing group than in the multi-lambing group,and in the uterus was significantly higher in the single lambing group than in the multi-lambing group,and there was no significant difference among other tissues.The results of this experiment showed that the expression trend of BMP15 and GDF9 genes in the gonadal axis of single and multiple lambing groups of Qianbei Ma goat was generally consistent,with the highest expression in the ovary and the lowest in the uterus.These results indicate that these two genes play an important role in the reproductive performance of Qianbei Hemp goats,which can provide a theoretical basis for further research on the reproductive mechanism of BMP15 and GDF9 genes in goats.
