绵羊BMP2、BMP4和BMP7基因多态性与产羔数的关联分析

摘要(Abstract):

为探究BMP2、BMP4、BMP7基因多态性与绵羊产羔数之间的关联性,筛选与绵羊产羔数相关的分子标记,本研究利用飞行质谱技术对5个群体(杜泊羊、滩寒杂交羊、杂一代、杂二代和横交一代)768只绵羊BMP2基因g.48462350C>T位点和BMP4基因g.63454744T>G位点以及BMP7基因g.58171856C>G位点进行检测,并与产羔数进行关联分析。结果表明:BMP2基因g.48462350C>T位点检测出CC、CT、TT3种基因型,在5个绵羊群体中均为中度多态(0.25G位点检测出TT、TG、GG3种基因型,在5个绵羊群体均表现为低度多态(PIC<0.25);BMP7基因g.58171856C>G位点检测出CC、CG、GG 3种基因型,在5个绵羊群体均为低度多态(PIC<0.25)。BMP2基因g.48462350C>T位点在杜泊羊群体中不处于哈代-温伯格平衡状态(P<0.05),在其他群体中均处于哈代-温伯格平衡状态(P>0.05);BMP4基因g.63454744T>G位点和BMP7基因g.58171856C>G位点在5个绵羊群体均处于哈代-温伯格平衡状态(P>0.05)。产羔数关联分析表明,3个位点不同基因型在5个绵羊群体中的产羔数无显著差异。综上所述,BMP2基因g.48462350C>T位点和BMP4基因g.63454744T>G位点以及BMP7基因g.58171856C>G位点不适用于上述5个绵羊群体多羔性状的选育。
In order to explore the association between BMP2,BMP4,BMP7 gene polymorphisms and litter size of sheep,and to screen molecular markers related to litter size of sheep,flight mass spectrometry was used to analyze 5 populations(DU Po sheep,Tan Han hybrid sheep,F1 hybrid sheep,F1 hybrid sheep,F1 hybrid sheep and F1 hybrid sheep) The g.48462350c>T locus of BMP2 gene,g.63454744t>G locus of BMP4 gene and g.58171856c>G locus of BMP7 gene were detected in 768sheep,and their association with litter size was analyzed.The results showed that CC,CT and TT genotypes were detected at g.48462350c>T locus of BMP2 gene,which were moderately polymorphic in 5 sheep populations(0.25G locus of BMP4 gene,which showed low polymorphism in 5 sheep populations(PIC<0.25);CC,CG and GG genotypes were detected at g.58171856c>G locus of BMP7 gene,which were low polymorphism in 5 sheep populations(PIC<0.25).The g.48462350c>T locus of BMP2 gene was not in Hardy Weinberg equilibrium in Dorper sheep population(P<0.05),but was in Hardy Weinberg equilibrium in other populations(P>0.05);BMP4 gene g.63454744t>G and BMP7 gene g.58171856c>G were in Hardy Weinberg equilibrium(P>0.05).Association analysis of litter size showed that there was no significant difference in litter size among the five sheep populations among the three genotypes(P>0.05).In conclusion,the g.48462350c>T locus of BMP2 gene,g.63454744t > G locus of BMP4gene and g.58171856c>G locus of BMP7 gene were not suitable for breeding multiple lambing traits of the above five sheep populations.
